The Expression Level of Retinol-Binding Protein in Peripheral Blood of Patients with Acute Myeloid Leukemia
Abstract: Objective To explore the expression level of retinol-binding protein (RBP) in peripheral blood of patients with acute myeloid leukemia (AML) and its relationships with clinical characteristics of the patients. Methods The expression level of RBP in patients with AML, those with benign hematopathy, and healthy subjects was analyzed. Further, the relationships between age, gender, cytogenetics, French-American-British (FAB) subtypes, molecular mutations, and RBP expression level were analyzed. Results Compared with AML patients, RBP expression level was significantly higher in healthy subjects and those with benign hematopathy. A significantly negative association was observed in patients with M (r=-0.499, P=0.036) and M (r=-0.412, P= 4 5 0.029). Acute promyelocytic leukemia (APL) patients showed higher RBP level compared with those of M (u=-2.085, P=0.037), M 2 4 (u=-3.085, P=0.002), and M (u=-2.249, P=0.024). However, no difference was observed between other FAB subtypes (all P > 0.05). 5 Elderly AML patients had higher RBP level compared with children (t=2.284, P=0.049) and adult patients (t=2.572, P=0.011). Patients carring FLT3-ITD mutation presented with lower RBP level than those without (u=-2.173, P=0.030). Conclusion RBP is not only a nutritional index for AML, but its expression level and relationships with clinical characteristics of AML might offer beneficial information for optimizing AML treatment.

Different Growth Hormone Receptor Expression in Human Gastric Carcinoma and the Proliferative Effects of Recombinant Human Growth Hormone
Abstract: Objective To knock down growth hormone receptor(GHR) in MGC-803 cells using vector-based small interference GHR (siGHR), and thereby to investigate the effects of recombinant human growth hormone(rhGH) on tumor growth and JAK2/STAT3 pathway of human gastric carcinoma xenografts with different expression states of GHR in nude mice. Methods Seventy-two male nude mice were bought for the experiment. Western-blotting method was used to detect the expressions of growth hormone receptor(GHR) in parental and transfected human gastric carcinoma cell lines MGC803. The gastric carcinoma cell lines of different GHR expression were then injected subcutaneously into 72 nude mice to create the subcutaneous xenografts models. These models were then randomly divided into three experimental groups: the control group(0.9%NaCl,0.2ml/d), the low-dose rhGH group(1U/(kg·d), 0.2ml/d) and the high-dose rhGH group(3U/(kg·d), 0.2ml/d). The mice in each group were all treated with different doses of drugs for 21 days continuously. The changes of body weights and tumor volumes of nude mice in each group were recorded. VEGF protein expressions in tumor tissues were detected by immunohistochemical method. Western Blot was performed to detect the components of JAK2/STAT3 signaling pathway. Results There is no significance of GHR expression in protein level between MGC-803 and MGC803-NC pGPU6/GFP/Neo-scramble-transfected cells(replaced with)(P>0.05). Compared to MGC-803 the GHR expression in pGPU6/GFP/Neo-shGHR-transfected cells(replaced with MGC803-shGHR) decreased significantly(protein decreased 57%). For parental and MGC803-NC groups, after giving rhGH for 21 days, the tumor volumes were significantly larger in the 3.0U/(kg·d) rhGH group than in the 1.0U/(kg·d) rhGH group and the control group(P<0.05). No significant difference was found in body weights of the nude mice among these 3 groups(P>0.05). Moderate positive staining with VEGF was observed in thecontrol group, while VEGF staining was strongly seen in rhGH administration groups. The expressions of p-JAK2 and p-STAT3 proteins for the 3.0U/(kg·d) rhGH group were both significantly higher than the 1.0U/(kg·d) rhGH group and the control group. The total JAK2 and STAT3 expressions of the 3 groups did not differ significantly(P>0.05). While for MGC803-shGHR groups, the body weights of the nude mice of 3.0U/(kg·d) rhGH group were much heavier than the 1.0U/(kg·d) rhGH group and the control group, but no obvious increasing effects on tumor growth and the expressions of VEGF, p-JAK2 and p-STAT3 were observed(P>0.05).Conclusions Recombinant human growth hormone can promote the growth of tumor in the MGC-803 and MGC803-NC xenograft tumor models in which GHR is highly expressed, but not for the MGC803-shGHR xenograft tumor models in which the expression of GHR is deeply decreased. Down-regulation of components of the expression of VEGF and JAK2/STAT3 signaling pathway may be the potential mechanisms.

Serum TC,HDL,apoA-I Levels on the Prognosis of Postoperative Sepsis in Cancer Patients
Abstract: Objective To observe the relationship between the level of total cholesterol (TC),high-density lipoprotein (HDL),apolipoprotein A-I (apoA-I) and the severity of postoperative sepsis in cancer patients. Methods This is a retrospective study. A total of 37 patients hospitalized in the surgery intensive care unit of the Third Affiliated Hospital of Sun Yat-Sen University from December 2010 to December 2011were enrolled in the study. All patients were divided into two groups according to the status after 28 days. The correlations between the lipoprotein and inflammatory cytokines, the independent predictive factor for sepsis were analyzied. Receiver-operating-characteristic curve (ROC) was used to evaluate the cut-off of TC, HDL, apoA-1 and APACHⅡas well as their predictive value for 28 days mortality. Results There was negative correlation between HDL, TC, apoA-1 and tumor necrosis factor-α(TNF-α)、 interleukin (IL-6) seperately; whereas these lipoprotein indexes didn't have significant correlation with neutrophil.The cut-off point of TC,HDL,apoA-I,APACHEII score were 1.46mmol/L,0.175mmol/L,0.525g/L,20.5 points. apoA-I has the highest diagnostic and predictive value. Conclusions The levels of TC, HDL, apoA-I can reflect the degree of inflammatory response to the postoperative sepsis and the severity of the disease in cancer patients. TC, HDL, apoA-I, APACHEII within 24hrs of sepsis have predictive value. But apoA-I was the highest diagnostic one.

Can Dietary Fish Intake Prevent Cancer?
Abstract: Objective The incidence of cancer showed an increasing trend in China in recent years. The development of cancer is closely related with diet, while fish is the main food source for humans. The aim of this study was to achieve a comprehensive assessment of the relationship between fish consumption and cancers risk. Methods Studies about fish intake and cancer risk, especially meta analysis were recruited and intensively summarized. Results Higher fish intake is associated with lower risk of lung cancer and colorectal cancer, but the associations of fish with risk of other cancers is controversial and inconclusive.Conclusions More well-designed epidemiological studies are warranted to further verify the effects of fish consumption on risk of various cancers.
