Leaky gut:mechanisms and clinical implications |

Abstract A contiguous layer of cells as well as the junctions that seal the paracellular space play a fundamental function to separate luminal harmful contents from the interstitium. Leaky gut resulted from impaired mechanical barrier and dysfunctional paracellular pathway, leading to increased intestinal permeability. Leaky gut has been proposed to be associated with a number of disease states, but there is a lack of valid clinical data supporting such proposal. Therefore, it is important to provide a balanced view of the association between leaky gut and diseases to facilitate the diagnosis and treatment of disease associated with increased intestinal permeability. This review summarizes the mechanism and clinical significance of leaky gut from aspects including the relationship between mechanical intestinal barrier and intestinal permeability, measurement of intestinal permeability, and intestinal permeability in disease states.
